从南北极环境样品中分离到7株产脂肪酶的细菌,经16SrDNA序列分析表明,这些细菌分别属于假交替单胞菌属(Pseudoalteromonas)和嗜冷杆菌属(Psychrobacter).采用p-NPP法对这7株细菌所产的脂肪酶进行酶学性质研究,结果显示这些酶的最适作用温度在30~40cC、最适作用pH值在7—8之间,在低温下能保持较高的剩余活力,对热敏感,属于适冷脂肪酶.其中假交替单胞菌(Psychrobacter sp.7342)所产脂肪酶具有低温下酶的剩余活力高、有效作用温度和pH范围广、热稳定性较好及对多种金属离子抗性强等特点.该菌株能利用多种单一氮源和碳源产酶,最适产酶温度为25℃.在此基础上进行正交实验分析得到了该菌株的最佳发酵条件为:蛋白胨和淀粉含量各为1.33%,酵母膏含量为0.3%,温度为25℃.  相似文献

The ocean plays a major role in global biogeochemical carbon cycling; it holds an important reservoir of reduced organic carbon, mostly in the form of dissolved organic carbon (DOC), and processes about one-half of the total primary production of the planet. Dissolved molecules present between living and assimilable size extremes (∼1000 nm-1 nm), constitute the most abundant form of remnant biochemicals in the ocean, outweighing the total living biomass by a factor of roughly 200. Because DOC is the fundamental substrate for marine microorganisms, and is primarily composed by small refractory biopolymers, this prompted the idea that the ocean might function as a huge repository of recalcitrant carbon. The missing link that elucidates this paradox and explains how the rich and vast stock of DOC becomes available to bacteria was the discovery that DOC throughout the water column remains in reversible assembly/dispersion equilibrium forming porous microscopic gels (Chin et al., Nature 391, 568-572, 1998). This abiotic DOC-POM shunt yields a microgel pool containing ∼70 gigatons of carbon forming discrete patches of high nutrient concentration that can be readily colonized by microorganisms. The presence of this huge gel mass in seawater extending far into the dark ocean has ramifications that might well scale nonlinearly through the microbial loop to the World Ocean and global climate system and it is fundamentally changing how oceanographers think about processes linking the microbial loop and biological pump to the rest of the biosphere and the geosphere. Even if a small fraction of DOC remains self-assembled, marine scientists will have to revise the rationale of established aquatic paradigms ranging from trace metal chelation, size–reactivity relationships, the microbial loop, the biological pump, colloid pumping, and humification. A ubiquitous, reversible DOC assembly/dispersion process implies a dynamic “patchiness” spanning from the molecular to the micron scale, where the assignment of static bulk features including dimension, concentration, functionalities and vertical fluxes can be open to question. This brief revision illustrates two case studies that show how simple methods and principles of polymer networks theory can be used to advance the understanding of one of the most intriguing and significant processes taking place in the ocean. Namely, the kinetics and thermodynamics of: (a) Ca-driven DOC self-assembly, and (b) hydrophobic bond-driven self assembly of DOC by amphiphilic exopolymers released by marine bacteria.  相似文献

以褐菖鲉(Sebastiscus marmoratus)为实验材料,设定苯并芘[B(a)P]的注射剂量梯度分别为0.00、0.05、0.50、5.00 mg/kg,分别于实验的第0、3、6天对每一条鱼进行腹腔注射,于第1、4、7天取样,测定胆汁中的B(a)P及其代谢物含量.采用荧光检测法检测B(a)P染毒后褐菖鲉肝细胞DNA-DNA交联(DDC)交联系数的变化,并用KC l-SDS沉淀法检测DNA蛋-白质交联(DPC)交联系数的变化.结果表明:(1)经注射染毒后鱼体胆汁中的B(a)P及其代谢物含量蓄积程度存在着明显的剂量效应和时间效应;(2)B(a)P的注射剂量分别为0.05、0.50 mg/kg时各时间点褐菖鲉肝细胞DDC交联系数均无显著增大(p〉0.05),在5.00 mg/kg剂量组,随着时间延长DDC交联系数逐渐增大,第4天即有显著差异(p〈0.05),第7天差异极显著(p〈0.01);(3)B(a)P诱导DPC的趋势与DDC类似,但0.5 mg/kg剂量组在第7天DPC交联系数即出现显著增大(p〈0.05).这表明B(a)P对DPC的诱导高于DDC.这为进一步研究B(a)P的遗传毒性提供了依据,并为探讨B(a)P致癌的作用机理提供了新的信息.  相似文献

The origin of perylene, a five-ring polycyclic aromatic hydrocarbon (PAH), in sediments has been a matter of constant debate and its formation mechanisms remain unclear. A perylene record in sediment was described and could support a clear link between perylene and terrigenous organic matter (OM) input. The distributions of PAHs, the variations of organic proxies such as the ratio of terrigenous to aquatic n-alkanes (TAR(HC)) and 5α(H)-stanols/Δ5-sterols ratio in sediments have been investigated. Sediments were sampled from a pond artificially created during the Middle Ages from a swampy area. In the period prior to the pond creation, for which high contributions of terrigenous OM were evidenced, perylene was found to be the predominant PAH. Furthermore, perylene content was shown to increase in response to the establishment of more reducing conditions. This result supports the common idea that the main source of perylene is natural and derives from biogenic precursors under reducing conditions. The creation of the pond in this wetland is marked by the deposition of a wood rich level characterized both by more oxygenated conditions and higher concentrations of perylene. These high concentrations could result from the introduction of high concentrations of biogenic precursors of perylene under oxidative conditions. Subsequently, the progressive burial of the woody level could have allowed the establishment of oxygen depleted conditions and the formation of perylene. These results validate the use of perylene as a paleoenvironmental marker of terrigenous sources but it must be carefully used as a marker for syn- or post-depositional oxygen depleted conditions in lacustrine environments.  相似文献

In coastal environments, fine-grain sediments often aggregate into large and porous flocs. ElectroMagnetic Current Meters (EMCM) and Laser In Situ Scattering and Transmissometry (LISST-ST) have been deployed within a Spartina alterniflora marsh of the Luoyuan Bay in Fujian Province, China, to measure the current velocity, the floc size and the settling velocity between 15 and 22 January 2008. During the observations, the near-bed water was collected in order to obtain the suspended sediment concentration (SSC) and constituent grain size. Data show that: (1) the nearbed current velocities vary from 0.1 to 5.6 cm/s in the central Spartina alterniflora marsh and 0.1–12.5 cm/s at the edge; (2) the SSCs vary from 47 to 188 mg/dm 3 . The mean grain size of constituent grains varies from 7.0 to 9.6 μm, and the mean floc sizes (MFS) vary from 30.4 to 69.4 μm. The relationship between the mean floc size and settling velocity can be described as: w s =ad b , in which w s is the floc settling velocity (mm/s), a and b are coefficients. The floc settling velocity varies from 0.17 to 0.32 mm/s, with a mean value of 0.26 mm/s, and the floc settling velocity during the flood tide is higher than that during the ebb tide. The current velocity and the SSC are the main factors controlling the flocculation processes and the floc settling velocity.  相似文献

用锰纤维富集-射气法测定了北黄海海水中的镭同位素226Ra和228Ra,研究了该海域水体中镭同位素的含量和分布.研究结果表明北黄海水体夏季226Ra的比活度为1.80~4.35 Bq/m3,平均值为3.06 Bq/m3;冬季226Ra的比活度为2.08~5.20 Bq/m3,平均值为3.28 Bq/m3.北黄海夏季228Ra的比活度为3.85~25.60 Bq/m3,平均值为10.60 Bq/m3;冬季228Ra的比活度为3.14~15.60Bq/m3,平均值为7.66 Bq/m3.该数据范围和中国近海其他海域、孟加拉湾、泰国昭披耶河口、濑户内海等海域相近.北黄海东北部海域,渤海海峡靠近山东半岛的海区和中北部海区表层镭同位素活度较高.C1断面镭同位素的分布特征从镭同位素的方面证实了渤海海峡水交换表现为北进南出特征这一结论的正确性.226Ra和228Ra的垂直分布较为复杂,大部分站位呈现出底层活度变高的趋势,其他少数站位呈现出中间层活度高的分布特征,不同来源的镭同位素输入至该海域形成了这样的分布特征.  相似文献

沼泽湿地土壤是氮的重要储库,发挥着源、汇和转化器的重要功能。湿地土壤中无机氮的物理运移不但影响着植物的养分供给状况,而且其对于湿地生态系统的结构、功能及健康状况等也有着深刻影响。综述了沼泽湿地土壤无机氮物理运移及影响因素的研究动态。当前湿地土壤无机氮物理运移的研究主要集中在硝态氮和铵态氮(特别是硝态氮)物理运移规律及部分影响因素(如水分条件、土壤物理性质等)的探讨上,缺乏无机氮物理运移的动力学、热力学机制与模型表征研究。鉴于当前研究中存在的问题,指出天然沼泽湿地是研究的薄弱点,其在今后应亟需加强的领域包括:①无机氮物理运移的驱动机制;②动力学、热力学与环境效应模型表征;③人类活动和全球变化对无机氮物理运移的影响。  相似文献

Effects of mixing on water mass subduction are analyzed in the South Pacific Ocean. Model simulations using a passive tracer and its adjoint are employed in conjunction with a particle tracking method to distinguish effects of mixing from those of advection. The results show that mixing processes can contribute to as much as 20% of the overall subduction rate in the South Pacific. Of this mixing contribution, about 30% can be attributed to meso-scale eddies, including their associated bolus transport, while the major part (70%) is due to other diabatic processes. The impact of mixing reaches its maximum near the Sub-Antarctic Front, accounting for nearly 30% of the total subduction rate. Consequently, estimates based on tracing particles or on advection alone may significantly underestimate the subduction rate in the South Pacific Ocean.  相似文献

In recent years,several studies pointed out that anthropogenic emission sources in China which significantly contribute to the PM2.5mass burden was an important cause of particulate pollution in South Korea.However,most studies generally focused upon a single pollution event.It is rare to see comprehensive research that captures those features prone to interannual variations concerning the transboundary pollutant contribution in South Korea using a unified method.In this paper,we establish the emission inventories covering East Asia in 2010,2015,and 2017,and then conduct the source apportionment by applying a coupled regional air quality model called the Integrated Source Apportionment Module(ISAM).Comparison of simulated and observed PM2.5mass concentration at 165 CNEMC(China National Environmental Monitoring Center)sites suggests that the PM2.5concentrations are well represented by the modeling system.The model is used to quantitatively investigate the contribution from emission sources in China to PM2.5concentrations over South Korea and those features found to be prone to interannual variations are then discussed.The results show that the average annual contribution of PM2.5has dropped significantly from 28.0%in 2010 to 15.7%in 2017,which strongly suggests that China has achieved remarkable results in the treatment of atmospheric particulates.  相似文献
